Projects

  • Masters Project - Process Economics: Whole bioprocess optimization for the manufacture of biosimilar mAbs

    -

The aim of the project is to extend the existing decisional tool (on a C# platform) with process economics and optimization features to address various challenges in multi-product facility design. Multiobjective optimization and multi-criteria decision making techniques are used to evaluate the tradeoff between process time and cost of goods for chromatography operations. Extension of the model will cover single use manufacturing, perfusion culture and continuous chromatography.

  • 3rd Year Design Project - Comparative analysis of Single-Use and Stainless Steel flowsheets for production of antibody fragment for treatment of colorectal cancer using E. Coli

    -

    - A process was defined to produce the chosen antibody fragment using E. Coli as the expression system for single-use and stainless steel equipment
    - Mass balances and process economics were subsequently evaluated
    - cGMP facility layout, familiarised with GMP guidelines
    - P&ID and mechanical drawing of a fermenter were produced using AUTOCAD

    Other creators
  • Analysis of Pichia fermentation results

    -

Laboratory work before term starts to analyse sorbitol content of samples taken throughout the fermentation performed previously and to analyse intracellular and extracellular protein before and after sonication of samples.

Skills: D-Sorbitol Colorimetric Assay, SDS-PAGE gel electrophoresis

    Other creators
  • Temperature characterization of a thermomixer for subsequent Fab' extraction from E. Coli

    -

The project aims to characterise an Eppendorf thermomixer before usage in a masters project for Fab' extraction in E. Coli.
Temperature profiles of different regions were analysed.

  • Effect of periplasmic heat extraction temperature and incubation time on Fab' purification

    -

The research project investigates the effect of periplasmic heat extraction temperature and incubation time on Fab' purification. Scale down heat extraction and chromatography experiments were performed on a robotic platform, involving PhyTips and HPLC experiments.
Further analytical work quantifying the product and contaminants involved SDS-PAGE gel electrophoresis, high throughput assays (BCA and Picogreen assays).

  • Industrial Challenges Associated with Pichia Pastoris Expression Systems: Do High Cell Densities Overburden Primary Recovery?

    -

A study on the behavior of Pichia pastoris under mixed (sorbitol and methanol) feed and pure methanol feed over course of fermentation. Pilot scale centrifugation was carried out on Baker’s yeast cell mixtures of various densities to mimic the various stages of upstream fermentation.
The project was led by UCL 2nd year PhD students:
Stefan Woodhouse, Alma Antemie and Ben Weil
Skills: P. pastoris fermentation, pilot scale centrifugation
